A collection of the ultimate no nonsense tough guy's from the games that I have played, just killing people doesn't count, you wouldn't really include Nathan Drake on a list like this would you? Would you?
I have probably made some obvious omissions, likely I have forgot, or I'm trying to bait people.

Anyone who has such a ridiculous title must be able to back up their claim, not only that but this is a name that the government awarded him! A man who is so tough that he is cloned with the sole intention of creating the perfect soldier, each of these clones could and probably should be this list but I'll reserve a sole spot for...
Not only he is a clone of the perfect soldier Big Boss but he actually surpasses him. He really is one of the few characters you fervently believe could get his way out of any situation. Even when he is seemingly in his death throws in MGS4 he get's the job done culminating in one of the most brutal fights I have ever witnessed in a game or film. I don't even think Snake needs any explaination for his inclusion, I assume most will see his name and nod knowingly.
A man of honour in a world of dishonour, Kazuma is a standout in the murky underworld and seemingly everyone wants a fight. He may try to diffuse the situation but if it comes to blows (It almost always does) he has no hesitation in dealing out his own brand of justice. He'll likely end a fight with the most unassuming inanimate object on hand, and make that look like the most brutal thing ever too. Even when he retires he is always brought back for the infamous 'one last job' and he still seems tougher than in the previous game.
He doesn't say much, he doesn't need too, he's tougher than you and he knows it. Even amongst an alien race known for their love of fighting he is esepcially strong, its no surprise that once he cuts loose after the events of the first game (I refuse to believe one bullet can stop him) he becomes the leader of his kind. His credentials speak for themselves when in Mass Effect 2 even after assembling the perfect squad to save the universe I still wanted to call on him above everyone else. Oh Wrex I have no doubt you would have laughed at that human sentinel, I did.
I'm not even the biggest Street Fighter fan but look at him! He even makes beads look menancing.
Angry is not the word, Kratos makes a career on the emotion. No matter the cause he'll mutilate anything that stands in his way, just because he can. Worst of all this chain of events is down to his own selfish desires. His shitlist consists of God's that he intends to kill. Not one to hold grudges because for the time that would take he's already riding towards you on the dissambled corpse of his previous victim.
The man's in shape, older than most of the fighers he faces but often comes out on top. Think's nothing of throwing his son off a cliff to see if he's worthy of his name and empire. After he suffers a similar fate he not only survives but claims back everything defeating his son Kazuya in the process. Not one for sharing, he would very much like to wipe out all of his family. Seemingly always being killed is not something which agrees with him and he always returns to claim whats his. He does all this in the midst of maintaining his trademark hair and tache combo, he has my respect.
Kills creatures by jumping on their heads, think about for a minute! Probably the most gruesome of all the way's to finish off a victim. "Has someone fallen on me? Oh no, wait, that's someone crushing my head through sheer force"
Cold. Calculated. Star of a mediocre action film. Agent 47 has seen it all, abides by his own moral code but will accept the job if it pays well. Can single handedly bring the Mafia to it's knees if the mood takes him. Even makes an enemy of God by leaving behind Religion.
He can carry on fighting with half a face missing, even more impressive is that it will regenerate so he can do it all over again the next time.
