Fountain of Youth or Agartha could be cool. Maybe they'll do some Japanese mythology. Have Drake chasing around the Three Sacred Treasures of Japan with weird stuff goin on around him.
What was the thing from the first one? Some cursed gold or coffin? I remember the totally-not-lickers but not exactly the story behind it. UC2 was the Cintamani stone and Shambhala and the Tree of Life all rolled into one, and the third had Iram of the Pillars and a vase that King Solomon had captured some less-than-kind jinn inside. So them having one thing doesn't necessarily mean it'll play out like you'd expect.
Or maybe they say fuck it and send Drake to The Lost World and it's just a really good Jurassic Park game. I'd play that.
All jokes aside, I think Libertatia going to be involved in this game judging by the teaser they released last year. That's a myth worth exploring. It will probably involved pirates, boats, and other related things popular in the mid-2010's.

It was El Dorado which turned out to not be a city of gold but a man of gold, specifically a coffin that contained a biovirus that the bad guys wanted. It's a really dumb game.

Hey, woah now! As realistic as the Indiana Jones trilogy? The very first one has a box that makes nazi faces melt. The reason why Uncharted always has some mythical element introduced at the end is specifically because of its Indiana Jones inspirations.

Yes, it is Mu.
The existence and final resting place of Excalibur/the sword in the Stone (different Swords at do the same thing.) Just as long as it isn't real...it would be way to easy to say "hey, because of Drake's Bloodline he was "worthy" this whole time!"
Atlantis would be fun. Lets them use the water physics stuff that they did for the boat section in Uncharted 3.

Well I've grown being told the legend and dreamt so much about Atlantis that it has a space in my imaginary, and I'd want to feel that wonder again.
But like you said it probably won't happen as it has already been done to death, and Naughty Dog has stayed away from the more popular myths.
The Garden of Eden might be a cool place to uncover. There are a variety of similar places in other religions, too so they wouldn't have to naturally go the Judeo-Christian route if they wanted to try something less "Indiana Jones".

If the teaser actually teased what the game will be about, pirates arre a' comin'. But I didn't see anything in it that linked to Libertatia, or was that just your assumption? I trust Naughty Dog to do something interesting, though they have lost a couple of good folks over the last years.

I'd actually like to see Agartha explored, I was quite dissapointed in The Secret World's portrayal of a hub world.

Personally I believe both are tied. The vase should contain a powerful hallucinogenic substance, and Talbot and Marlowe were both after it, to use in their shenanigans, as they both cultivate deceit as a weapon.

Indiana Jones gets pretty absurd for a movie that starts out that realistically. I mean, inflatable boat parachute, ripping out hearts, the lost ark, the holy grail, ALIENS. Yeah..

But it's this stuff that keeps Uncharted and Indiana Jones from just being a dude finding treasures. The super natural and mysticism is the best part.

They've always tried to take myths and put some pseudo-scientific twist on them. A curse actually being a virus; the cintamani stone actually being resin from a strange plant that dramatically alters biology; the jinns being the result of a strong hallucinogenic that came from the jar and seeped through into the water.
Come to think of it, it's all probably more grounded in "reality" than Indiana Jones.
I'd actually like them to pick something a little obscure. Though I knew of El Dorado and Shangri La prior to playing the series, it was enjoyable learning a bit about Iram of the Pillars through the course of the third game.
